Manamela has led the YCLSA since its launch in 2003. He was re-elected to the position of national secretary for a third term in December 2010. In addition, he serves on the politburo of the SACP, the party's highest decision-making body. Manamela is a member of the ANC's strategy committee in Parliament, which is responsible for the strategic planning of the ruling party's work in the National Assembly. He was appointed a whip of Parliament's committee on labour and he is a member of the Parliamentary committees on communications and on economic development. Mr Buti Manamela was the Deputy Minister in The Presidency from 26 May 2014 to 16 October 2017.

Mr Manamela completed an Electronic Engineering Honours Programme in Public Administration in 2008; he attended various Labour Courses with DHS ELA from 2002 to 2003; obtained a National Certificate, N5 between 1998 and 2001); and a National Senior Certificate in 1997.
